From Guaymas, buy a bus ticket (air conditioned) to Hermosillo. There you buy another ticket to Santa Ana, Caborda and Sonoyta, which is almost to the US border. At Sonoyta you buy a third ticket down to Puerto Penasco. If you're lucky, you can do this in one day. If not, take a sleeping bag or plan to get a hotel in Hermosillo or Caborca.
Alternatively, if you're accustomed to riding the not-airconditioned 'colectivo' buses (no head onboard), you might get one in Caborda that's taking the much shorter back road (Hwy 37) up to Puerto Penasco. We've driven it, and it's a pretty agricultrual area.
